Newgrounds Rumble[]
Alien Hominid[]
Alien Hominid is one of the primary targets for Piconjo's plan to destroy Newgrounds, as he is one of Newgrounds' heroes and protectors. Along with Pico, he is one of Piconjo's final opponents, and his demise leaves Newgrounds without any guardians, allowing Piconjo to decimate the Newgrounds universe.
In the first Newgrounds Rumble Challenge, "Tyrant Piconjo", Piconjo is terrorizing The Portal and submitting poorly made flash animations. Alien Hominid, along with Tankman and Pico, have been assigned to stop him. Alien Hominid fights against Convict again in the twelfth challenge "Royal Grumble Pt. 2", with the challenge having a heroes vs villains theme.

Alloy[]
In his plan to destroy Newgrounds, Piconjo discovers a new hero in the Newgrounds world named Alloy, who possesses of a powerful suit of armor. Piconjo decides to terminate him immediately to avoid disruptions to his plan.
In Alloy's story, he finds Tankman being attacked by Piconjo and Salad Fingers, and steps in to defend him.

Convict[]
While both Piconjo and Convict are Newgrounds' adversaries and desire its destruction, Piconjo is unwilling to share the credit for destroying Newgrounds with Convict. In Convict's story, he targets Convict when he learns of his plan, and attempts to kill him.
In the twelfth challenge, "Royal Grumble Pt. 2", Piconjo fights alongside him to defeat Pico and Alien Hominid.

Nene[]
After destroying P-Bot, Piconjo learns that Nene and Tankman are having an affair. As Nene and Tankman are two of Newgrounds heroes, he waits until they are alone, using this period as an opportunity to eliminate them both.

P-Bot[]
As Piconjo seeks to demolish Newgrounds, he is an adversary of those entrusted with defending it. In Piconjo's story, his first objective was to destroy P-Bot, who guards The Portal, the entrance to Newgrounds.
In P-Bot's story, he finds Piconjo trying to submit spam submissions through The Portal, and fights Piconjo to protect The Portal.

Pico[]
As Pico is the mascot and hero of Newgrounds, Pico is Piconjo's primary nemesis. Pico is one of Piconjo's final targets in his Story Mode, and his murder leaves Newgrounds without any protectors, allowing Piconjo to wreak havoc on the site.
In Pico's story mode, Piconjo is his final opponent. Pico is attempting to secure his position as the public face of Newgrounds, whereas Piconjo is trying to bring down Pico and assume control of the Newgrounds world.
Pico is dispatched to stop Piconjo when he threatens The Portal in the first Newgrounds Rumble challenge, "Tyrant Piconjo". Pico fights Piconjo again in the twelfth challenge "Royal Grumble Pt. 2", with the battle having a heroes against villains theme.

Salad Fingers[]
Though the exact reasons are unknown, Salad Fingers helps Piconjo attack Tankman in Alloy's story mode.
Samurai Asshole[]
Samurai Asshole is trying to find the one who destroyed his village. In his story, Piconjo is the first suspect he investigates, as he has the reputation of being a violent criminal. Samurai Asshole attacks him at Piconjo Manor.

Tankman[]
Because Tankman is a mascot for Newgrounds, he is an enemy to Piconjo. In Piconjo's story, he uncovers an intimate relationship between Nene and Tankman. When the couple believes they are alone, they maintain their relationship. This gives Piconjo the opportunity to murder them both.
In Alloy's story mode, Piconjo attacks Tankman once more, but Alloy intervenes to defend Tankman.
In Tankman's story, he attempts to become King of the Portal and to reign over Newgrounds. Piconjo is his final opponent. Knowing of Piconjo's reputation as a villain towards the people of Newgrounds, he attempts to kill Piconjo to increase his influence over the Newgrounds populace.
Tankman fights alongside Pico and Alien Hominid to stop Piconjo when he attacks The Portal in the first Newgrounds Rumble challenge, "Tyrant Piconjo".
